Some nearby premises were evacuated and residents in the area were advised to keep windows and doors closed and stay inside where possible.
The blaze was first reported shortly before 2am.
A Fire Service spokesman said there was no immediate threat to the public and there had been no injuries. But the building had suffered severe damage and a large amount of smoke was caused.
Traffic is still being disrupted, with long tailbacks in Knutsford Road, Chester Road, and Wilderspool Causeway.
Road closures are in force along Wilson Street from the Bridge Foot roundabout, up to Parker Street at its junction with Sankey Street.
Traffic from the south is being diverted across the Bridge Foot roundabout along the A49 towards Winwick Road.
It is thought these closures will remain for much of the morning and motorists are advised to avoid the area. A police helicopter was used to help control traffic.
The cause of the blaze is not yet known. But police have confirmed that a 43-year-old local man and two juveniles have been arrested in connection with the incident.
Police have appealed for information from the public and anyone who can help is asked to call thm on 101, quoting incident 63 of April 14.
Alternatively, information can be left anonymously on the Crimestoppers hotline 0800 555 111.
The club has been closed for several years and is earmarked to become a new “Youth Zone” providing a wide range of activities for young people.
What effect the fire may have on these proposals is not yet known.
